Abstract

Vitamin B12 is an essential cofactor for humans and is delivered to its two target enzymes via an intricate intracellular trafficking pathway. CblC is of the chaperones involved inB12 trafficking and converts cobalamin derivatives in the cytoplasm to a common cob(II)alamin intermediate. The cellular distribution of CblC is, as of now, inconclusive. Specifically, we want to determine whether CblC is located in the mitochondrion in addition to the cytoplasm. To address this question, we plan to use Western blotting, but an effective primary antibody to detect CblC is not commercially available. We have generated antibodies against purified CblC in chicken and purified the primary antibody and tested its efficiency in HEK293, HT29 cell lysates and in mouse liver. Upon completing the antibody characterization, we will separate the mitochondrial and cytoplasmic fractions from human cells to assess whether CblC is present in mitochondria.
