Abstract

Neuroendocrine Prostate Cancer (NEPC) is the most lethal form of prostate cancer. The prognosis and survival rate of patients with NEPC remains poor, and there are few viable therapy choices. Understanding the biology and molecular mechanisms that regulate NEPC development is critical for improving therapeutic outcomes. ESK981, a novel drug targeting the lipid kinase PIKfyve, has shown to be able to potentiate response to immune checkpoint blockade in castration-resistant prostate cancer (CRPC). This study aims to find whether ESK981 is efficacious in treating NEPC. The aims of this project are to evaluate the in-vitro inhibitory effect of ESK981 in the NEPC cell line NCI-H660; establish patient derived xenografts (PDXs) of NEPC with mouse models and the cell line implantation model; assess the in vivo efficacy of ESK981, along with the side effects; and determine the mechanisms of tumor growth inhibition of ESK981 in NEPC. We hope to provide a novel treatment option for NEPC patients and understand the mechanisms underlying the treatment.
