Abstract
Neutrophils and neutrophil extracellular traps (NETs) are critical mediators in thrombo-inflammatory diseases such as lupus and antiphospholipid syndrome (APS). Ginger root has traditionally been used as an herbal supplement in the treatment of various diseases. Gingerols are the most abundant compounds in ginger root, and we previously reported that administration of 6-gingerol to mice by intraperitoneal injection restrained neutrophil hyperactivity in models of lupus and APS. Here, we explored the impact of oral administration of Ginactiv® (Aurea Biolabs), a supplement that contains higher levels of gingerols than most available supplements (20% w/w versus 2%), on lupus/APS relevant large-vein thrombosis in mice. Administration of Ginactiv (150 mg/kg/day) orally for two weeks suppressed aPL-mediated thrombosis as indicated by clot weight (mean 6.64 vs. 3.8, p=0.0001). Importantly, levels of plasma NET were also markedly reduced in the same model (mean 0.73 vs. 0.55, p=0.002). This preclinical study is in support of our long-term goal to systematically study ginger supplementation and downstream effects in patients with APS and other autoimmune diseases.
