Abstract

This study focuses on BMPs, Bone Morphogenetic Proteins, in bone development and remodeling in mice. The effects of BMPs on bone growth and resorption have been widely studied, however synergistic outcomes with mechanosensing activity are not well known. The relevance of this study can help understand the mechanism of BMP signaling in bones when we exercise. This research can even help deepening our understanding in the field of craniofacial development. For our methodology, we had 6 groups of mice (n=10 per group) based on the combination of three conditions: age (either 6 or 9 weeks), suppression of Bmpr1a expression via a Cre-loxP technology that reduce BMP signaling, and exercise. CT scans, Raman spectroscopy, nano indentation, and dynamic histomorphometry were utilized for analysis of the cortical and trabecular bone development in the mice. We collected data on various related parameters, such as bone growth, crystallinity, hardness, etc. Though the results have not been finalized, disruption of BMP signaling actually decreases the bone formation rate, while disruption of BMP signaling has been found to increase bone mass due to further reduction of bone resorption. This tendency becomes more obvious when mice were exercised. Future data is still being analyzed about the cortical and trabecular bone characteristics. In terms of the future implications, alterations of BMP signaling can be applied and studied in humans. This can help direct us to treatment possibilities that prevent overexpression of BMP signaling.
