Abstract

The research that I conduct deals with understanding the physiological role of HPO-11, a protein which has been characterized as a pseudokinase. I am using C.elegans (worms)as my model organism to study the involvement of HPO11 in specific physiological processes such as vulval development and endosomal recycling in the intestine. Our readout is the GFP intensity of GFP-tagged proteins responsible for the above-mentioned physiological processes. I image these worms under two specific conditions : a. I knock down hpo11 expressions using RNA interference and b. I overexpress hpo11 in worm strains harboring GFP tagged proteins. I also look at Elongin B, a bonafide interactor and a potential target of HPO-11, to understand how these proteins interact. Overall, I use high-end imaging techniques followed by GFP intensity quantification to contribute to the limited knowledge about the function of HPO11.
