Abstract

Spinal Cord Injury (SCI) is currently very poorly understood and there is a lack of understanding about how the spinal cord responds to injury, as well as how it can be treated and repaired. Part of this is caused by the chronic inflammatory response, which prevents effective spinal cord cell repair. This study will look at previously collected data from researchers at the Shea Lab and utilize a new software, CellChat, in R programming to perform network communications analysis. SEURAT software was used for initial spinal cord sequencing analysis. Spinal cord cells and Spleen cells were analyzed in the data since they are the injury site and source of immune cells respectively. The spinal cord and spleen cells were then treated with nanoparticles and a PBS control solution separately. The preliminary studies for spleen cells indicated low relevant gene expression, and so they were omitted from the data analysis. We hypothesize that neutrophils will be the dominant signaling cells responsible for the inflammatory environment based on preliminary tests and their role in the immune system. Our results will utilize CellChat charts to provide more characterization on the signaling pathway connections between different cells. Overall, this research will be useful for future researchers who want to utilize CellChat in their research due to its unique network communications analysis as well as novel algorithms that can provide a new view on visualization signaling pathways. This could further help guide the study of spinal cord injury
