Abstract

This experiment aims to disprove the claim that SCD1 knockout (KO) adipocytes raised in colder conditions differentiate into beige adipocytes, and show that these KO adipocytes are actually undergoing cell death. Furthermore, this experiment focused on identifying the specific cell death pathway that these cells underwent by identifying cell death marker proteins that were present in the cells. SCD1 KO and control adipocytes were cultured and examined to see which types of cells survived and if they had died, which cell death pathway they went through (apoptosis, necrosis, or autophagy). Techniques such as immunoblot analysis and qPCR as well as visual examination under a microscope were used to identify cell death markers that were present in the SCD1 KO adipocytes. No final conclusions have been drawn yet, but it seems that autophagy is the most likely cell death pathway based on initial findings such as the presence of LC3 (microtubule-associated protein light chain 3) in these cells. If true, this information could have impacts in the pharmaceutical and medical fields since SCD1 is reported to induce several cancers.
