Abstract
Preliminary data from our rodent model of vertical sleeve gastrectomy (VSG) has sparked our investigation in the gut hormone, Fibroblast-Growth Factor 15/19 (mouse/human ortholog), which is known to pharmacologically regulate many aspects of energy homeostasis and glucose handling. This hormone is reduced in individuals with metabolic disorders and nonalcoholic fatty liver disease. Most importantly, postprandial plasma levels in humans and ileal FGF15 expression in mice increase after VSG. We show that intestinal-derived FGF15 is neccesary for the improvement in blood glucose regulation and to preserve muscle mass and bone mass after VSG-induced weight loss. These findings demonstrate a novel endocrine role for FGF15/19 in the regulation of multiple metabolic parameters, specifically in states of rapid weight loss. Although, weight loss is the desired outcome of bariatric surgery and weight loss strategies, the preservation of muscle mass and bone density are important parameters to be considered. The overall goal of the proposed studies is to test the hypothesis that FGF15/19 is necessary to maintain lean muscle mass, bone mass and glucose tolerance during rapid weigth loss by using preliclinical based studies.
