Abstract
Sepsis survivors are at an increased risk of long term death and rehospitalization. It is hypothesized that changes in the immune system mediate this risk. Our lab researches the long term effects on the immune system of patients after they have encountered sepsis. We use mouse models to evaluate long term changes in the immune system. A new and complete flow cytometry panel was developed to look at more than 20 cell types using cutting edge spectral flow cytometry in the lungs of both sepsis and non sepsis models. We calculated the staining index of 18+ antibodies in a flow panel on mouse lung. Analyzing these protocols allowed us to determine optimal antibody concentrations and then proceed with spectral flow cytometry and analysis. Research is still ongoing for this project, and completion of an entire panel with 18+ fluorophores in a mouse lung will be followed by evaluation of immune cells in the lungs of a naive mouse with that of lungs from a mouse surviving sepsis. The flow cytometry panel developed by this research will prove beneficial for scientists studying novel immune cells in the lungs.
