Abstract
It is well known that people respond to different diets in a variety of unpredictable ways. One of the most common diets seen in society today is the ketogenic diet. Our research will examine the effects of variation in food intake and weight gain while on a ketogenic diet. This will eventually be used to grow closer to our goal of using evidence to prescribe a diet to patients, and be confident in the predicted results. At the beginning of the project, we collected data from 47 diversity outbred mice both before and after four weeks of ketogenic diet treatment. We recorded things such as body weight, and amount of food consumed. After this phase of the experiment, we moved to other methods of quantitative data collection. We quantified the expression of appetite regulating hormones from the small intestines of the mice through procedures such as qPCR analyses of purified RNA. From this data we determined blood glucose levels, blood ketones levels, cholesterol, and triglyceride levels. We also measured the gene expression of six gut hormones present in the duodenum of the mice. We then analyzed and organized that data through R coding, working to build predictive models to draw conclusions from.
