Abstract
T-cell lymphoma is a type of cancer that forms in T lymphocytes. A case of peripheral T-cell lymphoma was sequenced through the Mi-OncoSeq clinical sequencing program at the Michigan Center for Translational Pathology (MCTP). Notably, mutations of the SOCS1 (Suppressor of Cytokine Signaling) gene and copy number alterations in the T-cell antigen receptor (TCR) loci were observed. An in-frame gene fusion between IL2 and TNFRSF17 was identified, using RNA-Seq. IL2 encodes a secreted cytokine, playing an important role in T and B cell proliferation and immune response. TNFRSF17 encodes a member of the TNF(tumor necrosis factor)-receptor superfamily, that is responsible for B cell development and associated with autoimmune responses. While TNFRSF17 is typically not expressed in T cells, its expression in the TCR loci was observed due to its fusion with IL2, inducing activity in oncogenic pathways. The IL2-TNFRSF17 fusion was cloned to create an expression construct. The activation of the IL2-TNFRSF17 fusion in the oncogenic NF-kappa B pathway was tested with protein assay and a Dual Luciferase assay, revealing insights into the development and progression of the lymphoma.
