Abstract
Understanding the mechanisms of age-related changes in cardiac physiology and impact on resilience to stressors such as ischemia are fundamental to develop strategies for healthier aging. We aimed to determine the impact of advanced aging (AA) caused by Hutchinson-Gilford Progeria Syndrome on the transcriptome of ventricular cardiomyocytes differentiated from human induced pluripotent stem cells (hiPSC-VCMS). hiPSCs from AA, a genetically related control (CTRL), and a CRISPR-corrected isogenic control (AAC) were submitted to ventricular cardiomyocyte-directed differentiation by manipulation of the WNT pathway with small molecules. hiPSC-VCMs were submitted to magnetic beads assisted purification and plated on MatrixPlus coated 6 well plates for 7 days for induction of maturation. hiPSC-VCMs were collected for total RNA extraction and submitted to the Advanced Genomics Core for polyA-enriched RNA sequencing. mRNA sequencing data was processed through the Bioinformatics core pipeline and iPathways was used for datamining and comparison between groups.
