Abstract
Hematopoietic¹ malignancies and leukemogenesis² are conditions of the blood correlated with decreased apoptosis³. While some of these conditions develop due to issues with cell cycle control or DNA damage, previous studies have also linked mutations in the epigenetic4 modifier SRCAP with these malignancies (Jahn, N., et al). Currently, the mechanism through which disruption of SRCAP function causes leukemia is unknown, but it is thought to have some effect on histone5 protein expression and/or function (Papaemmanuil, E., et al). The purpose of this study is to determine how SRCAP loss is related to the development of hematopoietic malignancies and whether this is through its regulation of histone expression or function. The goal is to understand how SRCAP functions epigenetically to potentially understand a new mode of epigenetic modification and how it may play a role in AML6 development. In this study, we performed various experiments with hematopoietic cell lines and mouse cells to determine how a downregulation of SRCAP leads to a fitness disadvantage. Additional cell lines and mutations were also studied to determine if SRCAP interacts with other genes in the process of regulating histone expression. 1.
