Abstract
The SWI/SNF chromatin remodeling complex plays a critical role in transcriptional regulation and is altered in over 20% of human cancers. Our lab has shown that targeting the SWI/SNF complex has been effective in treating transcription factor-addicted tumors like prostate and breast cancers. Most notably, recent work from our lab has also identified a subset of Ewing Sarcomas to be highly reliant on the SWI/SNF complex for survival. Ewing Sarcomas are a highly aggressive pediatric tumor subtype with few treatment options, highlighting the need to identify alternative therapeutic options. Using AU15330, a PROTAC degrader targeting the SWI/SNF ATPases, we show that about 8/19 (42%) Ewing Sarcoma cell lines are highly sensitive to loss of the SWI/SNF complex. Treatment of these lines with AU15330 causes significant cell growth inhibition and induction of apoptosis, as measured through c-PARP levels, providing an alternative therapeutic option for children with this lethal disease.
