Human microglia incorporation into self-organizing single-rosette human brain organoids

Abstract

Self-organizing single rosette brain organoid models can be used to model the neural tube and developing brain. The organoid models were developed from human induced pluripotent stem cells (iPSCs). The addition of relevant cell types, such as microglia, can be used to further develop the organoid into a more accurate model of the developing brain. In this experiment, human microglia were added at different stages and in varying amounts to the developing organoids. The microglia were added at multiple timepoints and in varying amounts in order to determine the combination of time and number that would best incorporate into the organoids and most accurately model the developing brain. When 1000 microglia were added to the organoids on day seven of the protocol, they were incorporated into the organoid. Additionally, the microglia continued to divide at a rapid rate. This model could be used in further research regarding the addition of other cell types to self-organizing single rosette brain organoids as well as providing a relevant model of the brain as a resource in future research.
