Abstract
Heart failure with preserved ejection fraction (HFpEF) is increasingly prevalent and lacks effective treatments due to its complex pathophysiology. This project evaluates the therapeutic potential of lysophosphatidic acid receptor 1 inhibition (LPAR1i) using both in vivo and in vitro HFpEF models. In mice, HFpEF was induced with a high-fat diet and L-NAME, followed by treatment with LPAR1i to assess its effects on cardiac remodeling. Cardiac function, blood pressure, and tissue hypertrophy were evaluated. In parallel, an in vitro HFpEF-like model was established by treating a cardiac cell line with angiotensin II and palmitate to mimic metabolic and hypertension stress. The results indicate that LPAR1 inhibition attenuates cardiac hypertrophy and tissue congestion without significantly affecting systemic blood pressure, suggesting a potential therapeutic role in limiting HFpEF-associated cardiac remodeling.
